It also covers a scientific study evaluating the effect of simulated microgravity on the activation of Rho that is involved in actin fiber remodeling in cells. The RhoA/Rho-kinase pathway may be a therapeutic target for treating PVR (pulmonary vascular resistance). The RhoA/Rho-kinase pathway is also involved in the pathogenesis of high flow induced pulmonary hypertension. For treating such disorders, Fasudil Hydrochloride (INN) has proofed to be a potent Rho-kinase inhibitor and vasodilator treatment able to lower pulmonary artery systolic pressure, suppressing pulmonary artery smooth muscle cells proliferation, attenuating pulmonary artery medial wall thickening and inhibiting right ventricular hypertrophy together with significant suppression of Rho kinase activity. Another drug, Atorvastatin, inhibits Rho signaling and is usually used for lowering blood cholesterol. Research, however, indicates that anti-RhoA small interfering RNA inhibits aggressive breast cancer more effectively than conventional blockers of Rho-mediated signaling pathways. This therapy was found to also be devoid of toxic effects.
